CompoundPorcine Cerebroside and Ganglioside Injection Attenuates Cerebral Ischemia/ReperfusionInjuries in Rat with Multiple Effects

Materials and methods:Adultmale Sprague-Dawley rats were subjected to MCAO surgery for 2 h and thenfollowed by reperfusion stage. All animals received treatment once a day for 14days from reperfusion started. Modified neurological severity score test wasperformed in 1, 3, 7 and 14 days after MCAO, and adhesive-removal test was performed only in 14 days after MCAO.Hematoxylin-eosinstraining was implemented to measure infarct volume and immunohistochemical experiment was performed to calculate thenumber of NeuN-positive neurons in ischemic cortex penumbra. Furthermore, theexpression of proteins associated with apoptosis (Bcl-2, Bax and GADD45α),synapse (Synaptophysin, SNAP25, Syntaxin and Complexin-1/2) and mitochondriafunction (KIFC2 and UCP3) were evaluated by western blot.
结论:
复方脑肽节苷脂对急性脑缺血再灌损伤大鼠具有较好的运动及感觉功能改善作用,可以从改善线粒体功能障碍、改善突触功能及抗氧化损伤等多方面发挥脑保护作用。而目前治疗脑缺血类疾病多从抗炎、溶栓、钙拮抗等方面来考虑,但是,相对于脑缺血再灌注复杂的损伤机制,简单控制其中一项或两项难以取得可靠全面的疗效,需从多方面考虑,运用多靶点协同治疗方案,以期达到最佳治疗效果。我们的研究结果显示复方脑肽节苷脂对脑缺血再灌损伤有多重保护作用,提示其具有强大的临床应用前景。
